During the election, President Barack?Obama crashed servers when he appeared in an impromptu "Ask Me Anything" post on Reddit.?Rep. Darrell Issa, a Republican from California,?decided to do the same ? "Together, we can make Washington take a break from messing w/ the Internet" his post said in part. But?the response wasn't quite as positive as he might have hoped.

Issa's post, seeking?discussion of a two-year ban on legislation that would affect the Internet, was submitted to?the popular social news site?Tuesday evening. Shortly afterwards, it reached the front page and questions started rolling in. By Wednesday morning,?the time Issa had set for answering questions, the queries?had turned into something of a dogpile.

By far the most popular question the one?posed by user The_Milkman: "Hey Darrell, why did you vote for CISPA?"?Indeed, Issa's sponsorship of CISPA???the controversial?Cyber Intelligence Sharing and Protection Act that was approved by the House, but has yet to be approved or vetoed by the president ????was the topic of many questions and much ire.

Issa's?attachment to the controversial piece of legislation?? which would expand?government agencies' ability to access data from sites like Facebook and Twitter???seemed to poison his attempt to solicit Reddit's help?from the start.

Regardless of the unfriendly atmosphere, Issa answered dozens of questions, including some openly hostile ones (his post record can be found here). And like the president, he included a picture of himself answering questions for authenticity's sake. Both the grilling Issa received and his responses could easily be considered more thorough than the president's, who answered questions some?commentators?characterized as softballs.

But regardless of the tone of the conversation, the goal of procuring constructive input on the proposed legislation (the "Internet American Moratorium Act" or IAMA)?seems to have been achieved. A number of comments and suggested changes have been submitted at the site set up for this purpose, and various issues have been raised in the comment threads that may help improve the bill. And the high positive vote count on the AMA itself indicates that the community is at least interested in discussing the topic.

There's a lesson to be learned for politicians and other?public figures who are considering this kind of open discussion on a site like Reddit: It's not always going to be a love-in, as it was with the president. You may end up like Issa, with a front-page post and 2,500 comments calling you everything from an opportunistic hypocrite to a crony of big business.

Amazon's supply chain 'more admired' than Apple rival

A global poll of more than 1,000 supply chain executives has revealed that Amazon?s supply chain is ?more admired? than Californian rival Apple on almost all major supply chain attributes.

The research, conducted by SCM World, asked 1,136 executives to rate the two companies on four supply chain attributes; agility, collaboration, execution and innovation. Amazon came out top on three of the four attributes, with Apple leading on innovation alone.

Over half of respondents (58 percent) said they admired Amazon most overall for the way it operates its Supply Chain, compared with 37 percent who picked Apple. Five percent of respondents said neither.

These survey results are a reversal of the findings by Gartner Supply Chain?s Top 25 for 2012, which ranked votes of 173 practitioners and considered companies? financial performance to place Apple first and Amazon second.

Kevin O'Marah, Head of Faculty at SCM World and co-author of a new report,?Apple and Amazon: Lessons for the Rest of Us, said: "The purpose of our research was to establish which of these two leading companies is most admired by the global supply chain community for the way it manages its supply chain, and to understand what lessons they have to teach other companies.

"The results show clearly that the majority of supply chain professionals believe that Amazon - and not Apple - is the master of supply chain excellence," added O'Marah, a former Group Vice President for Supply Chain at Gartner and co-creator, in 2004, of the (then AMR Research) Supply Chain Top 25 annual ranking.

Amazon and Apple are currently locked in a high-profile battle in the fast-growing market for tablet devices and the digital content consumed on them. The companies' rival products, the Kindle Fire and iPad, are expected to be among the hottest purchases this Christmas. ?

Stats breakdown

On?agility?(defined as the ability to quickly and cost-effectively shift amounts and/or types of production and delivery to improve operational performance in volatile conditions),?62%?of practitioners in SCM World's poll voted for Amazon, compared with?33%who picked Apple.

On?collaboration?(the ability to work across organisational boundaries to solve systemic operational problems and create new value for both customers and partners),?59%?chose Amazon, versus?31%?for Apple.

And on?execution?(the consistent and reliable delivery against commitments and within budgeted expenses), practitioners backed Amazon by?57%?to?38%.

However, Apple won hands down on the fourth key characteristic of supply chain excellence -?innovation?- taking a massive 78%?of the vote, compared with just?19%?for Amazon.

NFL Power Rankings 2012 Week 13: Skidding Vikings losing ground

The Minnesota Vikings are no longer being viewed as a top 15 team in the league following a disastrous loss to Chicago in Week 12.

Losers of three of their last four games, the Minnesota Vikings continue their mid-season slide down the power rankings of a number of top pundits from around the web.

Despite starting the year off at 4-1, Minnesota has fallen back to Earth in recent weeks to reach a 6-5 mark. SB Nation's Joel Thorman believes part of this has to do with the fact that the Vikings did not play Chicago or Green Bay earlier in the season, forcing him to drop the Vikings down two spots to No. 17 overall in his latest rankings.

CBSSports.com's Pete Prisco puts some of the blame on quarterback Christian Ponder. Prisco gave the Vikings the No. 17 spot in his rankings this week, but believes it's up to Minnesota's star quarterback to get this team back on track. "The fast start is being done in by an inability to throw the ball down the field," he said. "Christian Ponder has to do more."

Though they dropped down two spots from No. 14, the Vikings still managed to come in at No. 16 this week according to ESPN.com's updated rankings. The Vikings are ranked one spot behind a 5-6 New Orleans team, and exactly seven spots away from a 7-4 Green Bay Packers squad that sits just one game ahead in the standings.

French court delays Strauss-Kahn charge ruling

PARIS (AP) ? A French court on Wednesday pushed back a ruling on whether to drop aggravated pimping charges against Dominique Strauss-Kahn, the former IMF chief who has successfully fought accusations linked to his "libertine" lifestyle.

Three Appeals Court judges in the northern city of Douai set a new date of Dec. 19 to decide the demand by Strauss-Kahn to annul the charges linking him to a suspected prostitution ring run out of a luxury hotel in Lille.

Lawyers for Strauss-Kahn have claimed the investigating judges were biased.

The economist admits attending "libertine" gatherings but denies knowing that some women present were paid.

In August, a separate case against Strauss-Kahn centered on allegations of rape in a Washington D.C. hotel was dropped after prosecutors said the supposed victim, an escort girl, changed her account to say no rape was involved in the encounter when Strauss-Kahn headed the International Monetary Fund.

Criminal charges also were dropped by New York prosecutors in August 2011 in a case based on allegations of sexual assault by a hotel maid. But the high-profile case tarnished Strauss-Kahn's reputation. He resigned from his job as chief of the IMF.

If the judges decide to dismiss the aggravated pimping charges in December, Strauss-Kahn's legal load would be considerably eased ? but not fully gone. He would become an "assisting witness" in the case, meaning that if investigators discovered new elements implicating him he could face preliminary charges once again.

The prosecutors office has argued against dropping the charges, saying there are "serious" indications that justify the preliminary charges brought against Strauss-Kahn.

US defends 'enormous' climate efforts at UN talks

DOHA, Qatar (AP) ? Anticipating an onslaught of criticism from poor nations, the United States claimed "enormous" strides in reducing greenhouse emissions at the opening of U.N. climate talks Monday, despite failing to join other industrialized nations in committing to binding cuts.

The pre-emptive U.S. approach underscores one of the major showdowns expected at the two-week conference as China pushes developed countries to take an even greater role in tackling global warming.

Speaking for a coalition of developed nations known as the G77, China's delegate, Su Wei, said rich nations should become party to an extended Kyoto Protocol ? an emissions deal for some industrialized countries that the Americans long ago rejected ? or at least make "comparable mitigation commitments."

The United States rejected Kyoto because it didn't impose any binding commitments on major developing countries such as India and China, which is now the world's No. 1 carbon emitter.

American delegate Jonathan Pershing offered no new sweeteners to the poor countries, only reiterating what the United States has done to tackle global warming: investing heavily in clean energy, doubling fuel efficiency standards and reducing emissions from coal-fired power plants. Pershing also said the United States would not increase its earlier commitment of cutting emissions by 17 percent below 2005 levels by 2020. It is half way to that target.

"I would suggest those who don't follow what the U.S. is doing may not be informed of the scale and extent of the effort, but it's enormous," Pershing said.

"It doesn't mean enough is being done. It's clear the global community, and that includes us, has to do more if we are going to succeed at avoiding the damages projected in a warming world," Pershing added. "It is not to say we haven't acted. We have and we have acted with enormous urgency and singular purpose."

The battles between rich and poor nations have often undermined talks in the past decade and stymied efforts to reach a deal to keep global temperatures from rising more than 2 degrees C (3.6 F), compared to preindustrial times. Efforts taken in the absence of a deal to rein in emissions, reduce deforestation and promote clean technology are not getting the job done. A recent projection by the World Bank showed temperatures are expected to increase by up to 4 degrees C (7.2 F) by 2100.

Countries are hoping to build on the momentum of last year's talks in Durban, South Africa, where nearly 200 nations agreed to restart stalled negotiations with a deadline of 2015 to adopt a new treaty and extend Kyoto between five and eight years. The problem is that only the European Union and a handful of other nations ? which together account for less than 15 percent of global emissions ? are willing to commit to that.

Delegates in the Qatari capital of Doha are also hoping to raise billions of dollars to help developing countries adapt to a shifting climate.

"We owe it to our people, the global citizenry. We owe it to our children to give them a safer future than what they are currently facing," said South African Foreign Minister Maite Nkoana-Mashabane, who led last year's talks in Durban.

Environmentalists fear holding the talks in Qatar ? the world's biggest per capita emitter ? could slow progress. They argue that the Persian Gulf emirate has shown little interest in climate talks and has failed to reign in its lavish lifestyle and big-spending ways.

There was hope among activists that Qatar might use Monday's opening speech to set the tone of the conference. But Abdullah Bin Hamad Al-Attiyah, the president of the conference and a former Qatari oil minister, didn't offer any voluntary emission targets or climate funding for poor nations.

"Some countries, especially the one where we are sitting, have the potential to decrease their carbon emissions. They have the highest per capita emissions, so they can do a lot," said Wael Hmaidan, a Lebanese activist and director of the Climate Action Network.

"If nations that are poorer than Qatar, like India and Mexico, can make pledges to reduce their carbon emissions, then countries in the region, especially Qatar, should easily be able to do it. ... They still haven't proven they are serious about climate change."

Al-Attiyah defended Qatar's environmental record at a later news conference, insisting it was working to reduce emissions from gas flaring and its oil fields. Qatar is already doing plenty to help poor countries with financing, he said, adding that it was unfair to focus on per capita emissions.

"We should not concentrate on per capita. We should concentrate on the amount and quantity that each country produces individually," al-Attiyah said. "The quantity is the biggest challenge, not per capita."

The concentration of greenhouse gases such as carbon dioxide has jumped 20 percent since 2000, according to a U.N. report released last week. The report also showed that there is a growing gap between what governments are doing to curb emissions and what needs to be done to protect the world from potentially dangerous levels of warming.

At the same time, many scientists say extreme weather events, such as Hurricane Sandy's onslaught on the U.S. East Coast, will become more frequent as the Earth warms, although it is impossible to attribute any individual event to climate change. The rash of violent weather in the U.S., including widespread droughts and a record number of wildfires this summer, has again put climate change on the radar.

"While none of these individual events are necessarily because of climate change, they are certainly consistent with what we anticipate will happen in a warming world," Pershing said. "The combination of these events is certainly changing minds of Americans and making clear to people at home the consequences of increased growth in emissions."

In Washington, Rep. Ed Markey, D-Mass., urged the U.S. delegation at the talks to "heed the warnings from Sandy and other extreme weather supercharged by climate change."

"If the United States does not aggressively pursue sharp reductions in carbon pollution following the droughts, storms and other extreme weather events we have endured, the rest of the world will doubt our sincerity to address climate change," Markey said. "It's time to attack the carbon problem head on, and adapt to a climate already changed for the worse."

Many countries referenced Hurricane Sandy as a rallying cry for tough action to cap emissions, including a group of small island nations that said the monster storm may have jolted the world to recognize "that we are all in this together."

"When the tragedies occur far away from the media spotlight, they are too often ignored or forgotten," the island nations said in a statement.

Human Poop Helps Scientists Snoop on History

Human waste can map two millennia of history and climate change in a remote, Arctic settlement in Norway, according to a new study.

The findings suggest that human waste deposits could help researchers untangle the effects of natural and human-caused climate changes.

"We're able to really effectively disentangle what's human and what's natural," said study co-author Robert D'Anjou, a researcher at the University of Massachusetts. "We're able to date the onset of human settlement in the area and also look at agricultural practices and settlement history alongside the changing environment."

The report was published today (Nov. 26) in the journal Proceedings of the National Academy of Sciences.

Poop through the years

Archaeologists have used traces of ancient feces to recreate the history of specific archaeological sites: For instance, determining whether a latrine was ever used, D'Anjou told LiveScience. But nobody had used human waste to track the arc of human settlement and its effect on the environment. [Through the Years: A Gallery of the World's Toilets]

To do so, D'Anjou's team drilled several sediment cores from the bottom of the frigid Lake Liland on Vestv?g?ya Island, part of an archipelago of Norwegian islands north of the Arctic Circle. Since the Iron Age, people have farmed around this lake, and one of the largest Viking longhouses of the Arctic sits by the lake's frigid waters.

A 9.2-foot (2.8-meter)-long core captured about 7,000 years of time in the region, as well as 1.5-foot (45-centimeter)-long core containing sediments from about 2,300 years to 200 years ago.

The team analyzed coprostanol, a chemical component of human waste, as well as chemicals found in the waste of cows, sheep and other livestock. To tie human population levels to climatic changes, they also measured polycyclic aromatic hydrocarbons (PAH), a fingerprint of burning vegetation.

Levels of human and livestock waste byproducts jumped sharply around 2,300 years ago, when people first settled by the lake. At the same time, levels of PAH rose, probably because the new settlers burned forests to make way for grazing land and farming, D'Anjou said.

But from A.D. 650 to 850, human poop output dropped and the grassland gradually became reforested ? possibly because settlers left the shores of Lake Liland for the newly discovered territory of Iceland.

Another drop in human waste corresponded to the plague's peak during the late 1300s, when about 80 percent of the population moved from these areas or died off, D'Anjou said.

During the Little Ice Age, which lasted from the 1500s to the 1800s, PAH levels increased while the amount of coprostanol remained constant, indicating the settlers were probably burning much more wood to stay warm.

"In the Arctic, it's going to be very cold, so we see an increase in burning wood to keep warm," D'Anjou said.

Odd methods

The new study reveals how the climate and agriculture were intertwined, D'Anjou said.

"Slight variation in the growing season for agriculture greatly influenced the settlement and the population alongside this lake," D'Anjou said.

When the weather turned too cold for farming, settlers may have abandoned their farms for the coasts, where they fished for cod, he added.

But it also reveals a new way to piece together the history of the recent past, he said. Many times, researchers look only for specific compounds in sediments, but that limits their ability to recreate history, D'Anjou said.

"They have blinders on to what could be a really cool story in the strangest of locations," he said. "This one happened to come from poop."

Luck completed 20-of-37 passes for 240 yards on the day. Eight of those passes went to Reggie Wayne, who also drew a crucial pass interference penalty on Bills rookie corner Stephon Gilmore to help run out the clock at the end of the fourth quarter. Another one of Luck?s completions was to T.Y. Hilton for a touchdown in the third quarter, giving Hilton a touchdown catch to go with his 75-yard punt return score in the first half.

The touchdown drive in the third quarter featured Luck at his best as he completed six-of-nine passes and converted three third downs to keep the team rolling toward the end zone. Those moments are what lingers as the memory of Luck from the game. While they were less efficient than you?d like to see, he made enough of the big plays to keep drives alive to assure the Colts of a victory.

It wasn?t flashy, which also goes for the team?s defense. It was the best performance of the year for the unit, although it was short on takeaways (one interception which safety Tom Zbikowski fumbled right back to Buffalo) and other explosive plays. They gave up some plays that moved Buffalo down the field, but the red zone was kept under tight wraps on Sunday.

There aren?t any realistic playoff dreams left in Buffalo these days. C.J. Spiller ran for 107 yards on 14 carries, including a 41-yarder to set up Buffalo?s only touchdown of the day, but Ryan Fitzpatrick hit on just 17-of-33 passes for 180 yards to keep the team from taking advantage of several chances handed to them by a game defense. Mario Williams had three sacks, Justin Rogers had an interception and the Colts offense scored only 13 points. The Bills weren?t able to generate enough points, though, and that?s become a broken record in the wake of Bills games this season.

Online learning strategies of male and female students

ScienceDaily (Nov. 23, 2012) ? Research Highlight -- Virtual learning via cyber space. Researcher Rasaya Marimuthu and his team from Universiti Teknologi MARA conducted a study to determine the online learning strategies of male and female students in an English Language course.

Utilizing computers as a tool in education appears to be gaining momentum. There appears to be varying levels of interest in this method of online learning. This study attempted to investigate the relationship and differences (if any) of five variables, motivation, self monitoring, internet literacy, internet anxiety and concentration of students when engaging in online studying.

The study showed that there were no significant differences in the online learning experiences between male and female students. However, there existed correlations among some of the variables and also according to their gender. This study revealed that the respondents both male and female exhibited high levels of motivation, degree of self -monitoring, level of internet literacy and level of concentration when engaged in online learning.

As for the correlation effects of the five variables, the motivation in online learning had a significantly high positive correlation with self-monitoring in online learning. When comparison of the five variables according to gender was made, the results revealed that male and female students did not show any significant differences for the variables of motivation, self-monitoring, a higher level of internet literacy and also better levels of concentration when engaged in online learning.

The study appears to indicate that both male and female students in this study engaged in the same levels of motivation, self monitoring, internet literacy, internet anxiety and concentration in online studying. This study indicates that both male and female students are equally receptive to online learning.

This research was presented at the: 6th International Conference on University Teaching and Learning- Malaysia.

Small business Saturday | WPRI.com

PROVIDENCE, R.I. (WPRI) - After shoppers flooded store on black Friday, local stores in Rhode Island hope people shop local on small business Saturday.

For 12 years "Frod and Toad" on Hope St. has been selling everything from stationary to souvenirs.

Owner Asher Schofield tell Eyewitness News "People really come out for Small business Saturday. We see a huge jump in business. It's really a great thing. You really get a sense that people want to support their local mom and pop businesses."

According to the Small Business Administration, 100 million people nationwide shopped locally on small buisness Saturday last year.

"Why would we want to support a big store chain, they're going to make it anyway? so very much for the small business owner." said shopper Gail Goodwin-Gomez

Gail said she shopped last year and she plans to shop locally this year too.

"Luniac Glamour" on Main St. in East Greenwich opened just three months ago and is getting ready for it's first small business Saturday. The shop has all things beauty, from bags and jewlrey to hair and a nail salon.

Co-oweners Jaclyn Murphy and Kristen Marrah tell Eyewitness News that everything in their shop is made by Rhode Island designers.

"People who own the businesses are your friends and your family, people that you see on the street.// I think keeping everything local is ideal." says Kristen.

This is the third offical year of small business Saturday.

South Africa spent $3 billion on 2010 World Cup

(AP) ? South Africa spent more than $3 billion on the 2010 World Cup, and in return gained an "intangible legacy" from the first world soccer showpiece in Africa, the country's government said Friday in its final report on the tournament.

In the "2010 FIFA World Cup Country Report," released nearly 2? years after the event, South Africa's government said it spent $1.1 billion on building and upgrading stadiums alone.

Transport was the biggest cost, with $1.3 billion dedicated to improving road, rail and air links and a further $392 million on the country's main ports of entry.

In the absence of any final definitive figures on how much South Africa earned in total from being the host, the report said the World Cup had left an intangible legacy of pride and unity among South Africans and had changed the country's image as undeveloped, crime-ridden and dangerous in the eyes of the rest of the world.

"To top it all, we didn't have lions roaming the streets and we did have ATMs," the report, published by the ministry of sport, added light-heartedly.

It did predict a $6 billion boost to South Africa's economy as a result of the monthlong World Cup, according to a study by risk analysis and finance company Grant Thornton, but that was a mid to long-term projection.

FIFA reported it made a $631 million profit from the 2007-10 World Cup cycle and earned income of $3.65 billion from 2010 World Cup contracts. FIFA said it spent $1.298 billion on the World Cup in South Africa and also gave $100 million to the World Cup Legacy Trust, a fund that supports grassroots soccer projects.

"The World Cup in South Africa was a huge, huge financial success for Africa, for South Africa and for FIFA," FIFA president Sepp Blatter said in 2011 as the world body published its own financial report.

While critics have said that such a huge outlay on a 30-day sports event was impractical for South Africa ? and the final word on whether it was an economic success was still pending ? the government could argue that it had already earned over $400 million from the more than 300,000 tourists that visited for the World Cup.

The upgrade to much of South Africa's transport infrastructure was a long-term investment, the government said.

The expensive World Cup stadiums are still underused, however, and some are losing money. The Cape Town Stadium ? reportedly the most expensive of the seven new venues at $600 million ? is in the most trouble.

"This report will also serve as a reference guide and benchmark for planning other major sporting events," South Africa's government said in a statement to introduce the publication, which featured the 2010 World Cup logo and was embossed with shiny gold letters.

South Africa has said it is considering a bid to host the 2024 Summer Olympics.

France's Sarkozy: In court and political exile

PARIS (AP) ? He faces allegations he took advantage of an aging heiress to get envelopes stuffed with illegal cash for his presidential campaign. His party is mired in an internal feud. And still France's conservatives see Nicolas Sarkozy as their best hope to return to power.

It's a sign of how polarizing the former president is for the French: Many were suspicious of his close ties to the wealthy and threw him out of office; supporters see him as the only person able to save the country's economy and wish he'd return. On Thursday, the former president was in court to answer questions about whether he accepted illegal campaign donations from the 90-year-old L'Oreal heiress, Liliane Bettencourt.

Bettencourt's former accountant told police she handed over ?150,000 ($192,000) in cash she was told would be passed on to Sarkozy's campaign treasurer. In July, a magistrate ordered the seizure of Sarkozy's diaries, including his calendars.

The sum, although it pales in comparison to U.S. campaign funding amounts, shocked many French citizens because spending on political campaigns is tightly limited here. Individual campaign contributions to candidates are limited to ?4,600 ($5,930), and no candidate can spend more than ?22 million ($28 million) on an entire presidential campaign. The French government reimburses some of that money to the winner.

By comparison, an estimated total of $4 billion (?3 billion) was spent on campaigning for this year's U.S. presidential elections, and in the final months of the campaign each candidate was pulling in upward of $100 million (?77 million).

The investigation that has snarled Sarkozy centers on the finances of Bettencourt, Europe's richest woman and the focus of a long-running family feud over her fortune. Bettencourt, who was reported to suffer from Alzheimer's disease, has since been placed under legal protection.

Sarkozy's election loss in May cost him his immunity from prosecution. By July, his offices had been raided. Sarkozy's lawyer, Thierry Herzog, told the Sipa news agency that after Thursday's hearing Sarkozy could either face preliminary charges or be given special witness status with the possibility of facing charges later.

"Given the extremely strict rules and controls, illegal financing is completely impossible," said Claude Gueant, who was Sarkozy's campaign director at the time, chief of staff and later his interior minister.

Sarkozy entered the Bordeaux courthouse Thursday without notice after a decoy vehicle lured away the cameras hoping to catch him on the way in, according to the Sipa news agency. He and his lawyer met privately with the judge who will decide whether he faces charges.

Bettencourt's money was contested long before Sarkozy entered the picture. Her daughter accused a celebrity photographer of abusing her mother's alleged mental frailty ? and bilking the elder heiress out of ?1 billion ($1.3 billion) in cash and artworks. The elder Bettencourt's ex-butler leaked details of household conversations, including one in which the mother seems confused about whether or not she gave an island in the Seychelles to the photographer as a gift.

The island has since been sold off to a wildlife preservation organization. The photographer and an asset manager for Bettencourt face criminal charges of taking advantage of the L'Oreal heiress. And Bettencourt herself has been placed under legal protection of her relatives, including the daughter who first brought the case to light.

As the scandal grew, the French economy stalled, and French voters tossed Sarkozy and his conservative UMP party into the political wilderness. And now the internal vote to replace the ex-president as party leader has descended into bitter allegations of questionable vote counts, leaving the party in chaos.

A poll released Thursday showed that a majority among the conservative UMP party think Sarkozy would be the best presidential candidate for the 2017 elections ? nearly double the percentages of those who backed the two men now wrestling for power.

The person mediating the party dispute is Alain Juppe, a former prime minister who was convicted in 2004 in an illegal party financing scheme and has since bounced back as a power broker in the conservative party.

Former President Jacques Chirac, another conservative caught in that same scandal, is the only French leader since World War II-era Nazi collaborator Marshal Philippe Petain to be charged or convicted of a crime.

When conservation goes genomics: Finding needles in a haystack

Studying the genetic variability of endangered species is becoming increasingly necessary for species conservation and monitoring. But, endangered species are difficult to observe and sample, and typically harbour very limited genetic diversity. Until now, the process of finding genetic markers was time consuming and quite expensive. These obstacles make the collection of genetic data from endangered animals a difficult task to fulfill. A research team led by Loun?s Chikhi, group leader at the Instituto Gulbenkian de Ci?ncia (IGC) and CNRS researcher, has now contributed to change the odds when looking for diversity. Taking advantage of cutting edge DNA sequencing methodology and the collaborations with the Sabah Wildlife Department in Malaysia, Rachel O'Neill's laboratory and a private company, they were able to identify the genetic markers for the Bornean elephant, an endangered species, using blood from very few animals. The results showed that Bornean elephants have very low genetic variability that can impact on their survival to a threatened habitat, but that variable genetic markers can still be identified. The study now published in the journal PLOS ONE*, besides contributing to the conservation of the Bornean elephant, opens new avenues for the conservation of other endangered species.

The Bornean elephant is a unique subspecies of the Asian elephant, with a quite distinct morphology and behavior. They are generally smaller than other elephants, with straight tusks and a long tail. Currently, there are around 2000 individuals, located only in the North of Borneo. It remains unknown how this population of elephants evolved to become so different and why its distribution is so restricted.

Despite being one of the highest priority populations for Asian elephant conservation, until now there were limited genetic tools available to study its genetic variability and none that had been specifically designed for this species. Now, in the work conducted by Reeta Sharma, a Post-Doctoral fellow in Loun?s Chikhi's group, for the first time DNA sequences that characterize the genome of the Bornean elephants, called genetic markers, were identified. The research team used two different DNA sequencing technologies that are fast and increasingly cheaper. This kind of technology has been used for common laboratory species such as mice and fruit-flies, but they are only now starting to be used on endangered and "non-model" species.

Until now, in order to determine whether the species still harboured sufficient genetic diversity it was necessary to look through huge regions of the genome, using classical genetics methodologies, or use markers developed for other species, with varying levels of success. This approach can become unsustainable for the endangered species, whose numbers have gone bellow a certain size for long time. The only study that previously had tried to analyse Bornean elephants, using genetic markers developed for other Asian elephants had found nearly no genetic diversity. The work now developed demonstrates that if the methodology can be applied to the Bornean elephant, it should be possible to find the needles we need, and not get stuck with the hay, i.e., to find variable genetic markers in many other species.

The DNA analysis done resulted from blood samples collected only from seven Bornean elephants from the Lok Kawi Wildlife Park (Sabah, Malaysia) and from Chendra, the star elephant of Oregon zoo (Portland, USA). But, the research team is confident that these DNA sequencing methods can be used to type genetically other biological samples, such as hair or faeces, easier to obtain from wild animals, even though blood or tissue samples are still necessary to identify the markers during the first steps.

Reeta Sharma, first author of this work, says: 'The methodology applied to identify the genetic markers for the Bornean elephant can be used in the future for studies on the genetic variability of other species or populations facing the risk of extinction.'

The Bornean elephants live in an environment where natural habitats disappear quickly, due to oil palm plantations and populations get isolated from each other. Having access to variable genetic markers will be crucial to identify populations that are isolated and genetically depauperate, and monitor them in the future.

The origin of these elephants in Borneo raises controversy that has been long discussed. The only study done on the basis of genetic data concluded that they had been present in Borneo for more than 300,000 years. This theory does not satisfy all researchers as there is lack of elephant fossils in Borneo to support it.
